Transcriptome analysis of the effect of diosgenin on autoimmune thyroiditis in a rat model

Abstract In a mouse model of Graves’ disease (GD), diosgenin has been shown to have therapeutic effect on GD by alleviating goitre. However, research the autoimmune thyroiditis (AIT) is lacking. this study, transcriptomics was used comprehensively analyse protective against AIT in rats and possible mechanism. The results showed that diosgenin-intervention group, compared expression serum triiodothyronine, thyroxine, free thyroxine decreased thyroid-stimulating hormone increased; these changes were accompanied downregulation thyroglobulin, TSH receptor antibody thyroid peroxidase serum. Furthermore, transcriptome detection, RT-qPCR immunohistochemistry verification revealed tissue, relative mRNA protein cyclic adenosine 3?,5?-monophosphate (cAMP), kinase A (PKA) cAMP response element-binding (Creb) increased S100 calcium-binding A9 (S100A9) groups. summary, alleviates development AIT, possibly via activation cAMP/PKA/Creb pathway S100A9 gene expression.
